- Quantitative Estimation of Nevirapine by High Performance Thin Layer Chromatography

A simple, accurate and precise HPTLC method has been develop ed and validated for the estimation of nevirapine from bulk drug and tablet formulations. The s eparation was achieved on TLC plates using appropriate solvent system. The spots so devel oped were densometrically scanned at 283 nm. The linearity of the method was found to be wit hin the concentration range of 2.50μg/ml to 62.50μ/ml. The validation parameters, tested in ac cordance with the requirements of ICH guidelines, prove the suitability of this me thod. The method was successfully applied for determination of drug in tablets, wherein no interference from tablet excipients was observed, indicating the specificity of the developed method. Thus the proposed method can be used successfully for routine analysis of nevirapine from capsule and tablet formulations.Keywords

Stability testing is to provide evidence on how the quality of plant products varies with the time under influence of a variety of environmental factors such as temperature and humidity. Hence, kokum juice was subjected to accelerated stability studies and evaluated by anthocyanins content. Anthocyanins are present in the fruit of Garcinia indica, which is responsible for colour of kokum juice. The anthocyanins in juice were studied during six months period by subjecting it to accelerated conditions. The total acidity, brix and pH of the formulation were also monitored as the parameter of stability testing. The study was carried out as per the ICH guidelines.
The method used for total anthocyanins determination involved UV spectroscopy. Measurement of the absorbance was done at 510 nm on sample diluted with pH 1.0 and 4.5 buffers. The pigment content was calculated in absolute quantities with the aid of extinction coefficients. 30 ml of juice was withdrawn from four samples of capacity 200 ml each on 0, 1 and 3 month intervals. The results suggest that an increase in temperature decreases the anthocyanins stability. The degradation of anthocyanins in kokum juice samples was observed which is out of the limits of acceptance criteria according ICH guidelines.

The analytical industry today is showing a serious shift in the paradigm worldwide with advancements in technological compatibility and integration. Traditional analytical approaches including HPLC (High-Performance Liquid Chromatograph), GC (Gas Chromatograph), UV (Ultraviolet) detection, etc., have become insufficient to effectively address the growing number of challenges in analyses of species- specificity and sensitivity. This has given rise to investigate and offer highly advanced hyphenated techniques that meet the needs of high-technology oriented applications. Hyphenation-linking together of "standard" analytical techniques-generally leads to enhanced analytical performance. The biggest advantage of hyphenated speciation techniques is the ability to detect species other than the pre-conceived compounds. The advancement of analytical techniques is thus bringing a new era of development which will serve as a rapid and unambiguous tool in the drug development process.Keywords

A simple, specific, accurate, and precise reverse phase liquid chromatographic method (RP-HPLC) was developed and validated for the estimation of Metoprolol succinate from bulk drugs. A RP Spherisorb C-18 (Waters) 10μm column having 250×4.6 mm ID in isocratic mode, with mobile phase containing acetonitrile: methanol: 10 mM aqueous phosphate buffer (20:20:60%v/v) was used. The flow rate was 1.0 ml/min and effluents were monitored at 254 nm. The retention time of Metoprolol succinate was 5.1 min. The linearity of the method was good (r > 0.998), as also were intra-day and inter-day precision (RSD <2%).
The method was validated for accuracy, specificity, limit of quantification, limit of detection, robustness and stability. The results showed that proposed method is successfully applied for the quantitative determination of Metoprolol succinate in bulk drugs.
